No. It does not need you to be hooked up to city water while you are using it, if that is what you mean by running water.
But it does need a water supply from somewhere:

The normal install is to tap into your RVs fresh water system using the included 1/2 inch tee and 1/4 inch water line.

The cooler has a float valve that maintains water level in the pan as long as it has water pressure feeding it.
So in real life:

Boondocking: it will run off your fresh tank, and you will use your 12V pump to supply water.

On hookups: it can use city water (still through your RV plumbing).
One more thing: it consumes water as it cools (thats how evaporative coolers work), so your runtime off the fresh tank depends on how hard you run it and how dry the air is. It it ran 24 hours it would use roughly 3 gallons of water.
